چکیده مقاله:

The effect of aqueous smoke extract, derived from Tanacetum parthenium, onin vitro plantlet regeneration from rapeseed (Brassica napus L. cv. Topas)microspore-derived embryos (MDEs) was evaluated in this study. Inoculation ofrapeseed MDEs with the smoke-extract dilutions (1:250, 1:500, 1:1000 and 1:2000)for various times (5, 10 and 15 min) significantly enhanced the percentage ofregenerated plantlets from these embryos and also decreased the callogenesispercentage in these explants. The root length and shoot length of inoculated MDEswere significantly improved with all smoke-extract dilutions except for dilutions of1:100 and 1:10. Application of smoke-extract solutions (1:1000, 1:500, 1:250 and1:100) in B5 regeneration medium significantly enhanced the plantlet regenerationfrom rapeseed MDEs compared to control cultures. Furthermore, use of smokeextractsolutions in combination with GA3 (0.1 and 0.15 mgl-1) significantlyimproved the root length and shoot length of rapeseed MDEs compared to separateuse of smoke-extract solutions. Use of filter paper saturated with aqueous smokeextract (1:250) on top of the B5 regeneration medium was the best smoke treatmentmethod improving the plantlet regeneration percentage, root length and shootlength of rapeseed MDEs compared to other treatment methods. The conversionfrequency of rapeseed MDEs pretreated with smoke extract dilution of 1:500 wasimproved up to 50% compared with the control MDEs (26.67%), whereas the useof smoke extract (1:100) in B5 regeneration medium enhanced the conversionfrequency of these MDEs up to 62% compared with the control cultures (

Triacylglycerol Bioassembly in Microspore-Derived Embryos of Brassica napus L. cv Reston.

Erucic acid (22:1) was chosen as a marker to study triacylglycerol (TAG) biosynthesis in a Brassica napus L. cv Reston microspore-derived (MD) embryo culture system. TAGs accumulating during embryo development exhibited changes in acyl composition similar to those observed in developing zygotic embryos of the same cv, particularly with respect to erucic and eicosenoic acids. However, MD embryos...

Effects of gibberellin, abscisic acid and embryo desiccation on normal plantlet regeneration, secondary embryogenesis and callogenesis in microspore culture of Brassica napus L. cv. PF704

Most of the microspore-derived embryos can not regenerate normally in rapeseed. The effects of gibberellins (GA3), abscisic acid (ABA), and embryo desiccation on normal plantlet regeneration were studied. The donor plants were grown in a growth chamber at day/night temperatures of 15/10˚C with a 16/8h photoperiod, respectively. Microspores were isolated from whole buds of 2.5-3.5mm in length, c...

In vitro Shoot Regeneration Improvement with Thidiazuron and Benzyladenine in Rapeseed (Brassica napus L.)

An in vitro method was developed for high frequency shoot regeneration and continuous production of Brassica napus L. in large number. The different concentrations of benzyladenine (1.5, 3.0 and 4.5 mgl and thidiazuron (0, 0.15 and 0.30 mgl) were evaluated for shoot and root regeneration using hypocotyl explants with 7, 14 and 21 days old. Shoot regeneration (SR) was affected by both benzyladen...

Biosynthesis of Acyl Lipids Containing Very-Long Chain Fatty Acids in Microspore-Derived and Zygotic Embryos of Brassica napus L. cv Reston.

Biosynthesis of very long chain (>C(18)) fatty acids (VLCFAs) and the pathway for their incorporation into acyl lipids was studied in microspore-derived (MD) and zygotic embryos of Brassica napus L. cv Reston. In the presence of [1-(14)C]oleoyl-coenzyme A or [1-(14)C] eicosenoyl-coenzyme A, malonyl-coenzyme A, and reducing equivalents, maximal in vitro elongation activity was expressed in prote...

The Effect of Carbohydrate Source on the Development of Brassica Napus L. Immature Embryos in Vitro

The effect of fructose, glucose, maltose and sucrose at various concentrations on Brassica napus embryos was studied. The morphogenetic reaction depended on the type and concentration of sugar in the medium. The frequency of developing embryos was highest on sucrose, followed by maltose and glucose. Fructose did not stimulate embryo growth. Spectrophotometry of autoclaved fructose showed an abs...

The role of arabinogalactan proteins binding to Yariv reagents in the initiation, cell developmental fate, and maintenance of microspore embryogenesis in Brassica napus L. cv. Topas.

Arabinogalactan proteins (AGPs) are extracellular proteoglycans involved in plant growth and development. The addition of beta-D-glucosyl Yariv reagent (betaGlcY), a synthetic phenylglycoside that specifically reacts with AGPs, to the culture medium notably disturbed microspore embryogenesis in a concentration-dependent manner. The initiation of microspore embryogenesis was clearly inhibited by...
