چکیده مقاله:

The genus Tulipa L. (Liliaceae) comprises about 100 species and Iran is considered as one of the main origins of tulips. In this research, genetic diversity and population structure of 27 wild populations of tulips collected from Iran were studied by 15 highly polymorphic and reproducible expressed sequenced tag-simple sequence repeat (EST-SSR) markers and 8 nucleotide binding site (NBS)-enzyme combinations. According to EST-SSR genotyping, the average of gene diversity (GD) and polymorphism information content (PIC) were 0.66 and 0.62, respectively. However, the values of GD and PIC were equal for each NBS primer–enzyme combination which ranged between 0.85-0.95 with a mean value of 0.91. The mean value of resolving power (EST-SSR = 1.93; NBS-LRR = 17.39) indicated that the NBS markers had higher discriminatory power compared to the EST-SSR markers. UPGMA clustering confirmed the results of PCA which was further confirmed by Bayesian model-based STRUCTURE analysis. Population structure analysis detected 3 and 4 gene pools for 27 wild tulip germplasms with EST-SSR genotyping and NBS-LRR profiling, respectively. The AMOVA results indicated that molecular variation among populations (ΦPT = inter-population variation) was 82% and 93% of the total variation for EST-SSR and NBS-LRR markers, respectively. The results of this study will help the conservation and phylogenetic studies of tulips.

Genetic diversity of Iranian melon cultigens revealed by AFLP markers

Iran is a part of the secondary centre of origin of melons, and therefore, there is a wealth of genetic variation of these species in this country. The Amplified Fragment Length Polymorphism (AFLP) marker was applied to investigate the genetic variations among five major Iranian melon cultigens (Cucumis melo) and ‘Ananasi’ as a general well-known cultivar. Ten primer pairs were used on 90 indiv...

Genetic diversity and relationships among different tomato varieties revealed by EST-SSR markers.

The genetic diversity and relationship of 42 tomato varieties sourced from different geographic regions was examined with EST-SSR markers. The genetic diversity was between 0.18 and 0.77, with a mean of 0.49; the polymorphic information content ranged from 0.17 to 0.74, with a mean of 0.45. This indicates a fairly high degree of diversity among these tomato varieties. Based on the cluster analy...

Genetic population structure and differentiation of Western Iranian Oxynoemacheilus argyrogramma (Heckel, 1847) using SSR markers

This study was carried out to investigate the genetic diversity and population structure of 90 specimens of Oxynoemacheilus argyrogramma collected from Sepidbarg, Gamasiab and Ghaleji rivers, in the west of Iran. Analyses using three microsatellite loci indicated that the average number of alleles in the population was 12, which was well above the reported values for freshwater fishes. The expe...

Genetic variation, population structure and linkage disequilibrium in Switchgrass with ISSR, SCoT and EST-SSR markers

BACKGROUND To evaluate genetic variation, population structure, and the extent of linkage disequilibrium (LD), 134 switchgrass (Panicum virgatum L.) samples were analyzed with 51 markers, including 16 ISSRs, 20 SCoTs, and 15 EST-SSRs. RESULTS In this study, a high level of genetic variation was observed in the switchgrass samples and they had an average Nei's gene diversity index (H) of 0.311...

Assessment of Genetic Diversity and Population Genetic Structure of Corylus mandshurica in China Using SSR Markers

Corylus mandshurica, also known as pilose hazelnut, is an economically and ecologically important species in China. In this study, ten polymorphic simple sequence repeat (SSR) markers were applied to evaluate the genetic diversity and population structure of 348 C. mandshurica individuals among 12 populations in China. The SSR markers expressed a relatively high level of genetic diversity (Na =...
