P-59: Low IVF Rate and Embryonic Development in Ciprofloxacin-Treated Male Mice

Background: In the last few years, a marked decrease in male fertility has been reported. Environmental factors were recently suspected for this problem. The misuse of medicines especially antibiotics are among environmental factors that can affect reproductive system. Ciprofloxacin is a synthetic antibacterial agent belonging to the family of fluoroquinolones with a very broad spectrum effect against microbial pathogens. The aim of this study was to investigate the adverse effects of ciprofloxacin on ivf rate and embryonic development in male mice. Materials and Methods: Adult NMRI mice were randomized and kept on standard diet pellets and water adlibitum.ciprofloxacin was suspended in carboxymethyl cellulose(CMC) and given orally in doses of 206,412mg/kg body weight once daily for 45 consecutive days.CMC was given concurrently to control animals.At the end of the study the animals were sacrificed humanely and sperm processing for in vitro fertilization was performed,then sperms was added to HTF+BSAmedium containing the oocytes.All of the HTF droplets were covered with mineral oil.The rate of fertilization,two cell embryos,blastocysts,arrested embryos and type of them was examined in period of 120 hours. Results: A significant decrease in fertilization rate,percentage of two cell embryos and blastocysts were observed in ciprofloxacin - treated groups compared to the control group.Moreover administration of ciprofloxacin caused significant increase in embryo arrest rate. Conclusion: The present study showed that administration of ciprofloxacin decreased in vitro fertilization rate and percentage of two cell embryos and blastocysts.
