چکیده مقاله:

Among the two types of bacterial L-asparaginases, only type II enzymes have been used inthe treatment of acute lymphoblastic leukemia owing to their higher affinity for L-asparagine.However, current screening media used for the isolation of L-asparaginase-producingmicroorganisms do not discriminate between the two types of L-asparaginase. During anoptimization study conducted to increase L-asparaginase production by environmentalHalomonas isolates, it was noticed that the pattern of L-asparaginase production in responseto variations in glucose concentration varied between different isolates suggesting that theydiffer in their ability to produce type II L-asparaginases, an observation that was confirmed byfurther experiments. Bioinformatics analysis of available Halomonas whole genome sequencesrevealed that indeed some species of this genus possess both L-asparaginase types while otherspossess only type I enzymes. By comparing the growth pattern of these isolates on differentmedia, we propose that by omitting glucose, reducing the concentration of L-asparagine andproviding an alternative nitrogen source in L-asparaginase screening media it may be possibleto differentiate between type I and type II activities.

New targets for old diseases: lessons from mucolipidosis type II

Lysosomal storage disorders (LSDs) are inherited diseases characterized by progressive intracellular accumulation of undigested macromolecules within the cell due to specific lysosomal defects. Lysosomal storage results in a global impairment ofmany lysosome‐dependent pathways (e.g. autophagy and endocytosis), leading to cellular dysfunction and death (Ballabio &Gieselmann, 2009). LSD patients ...

Immunological studies on guinea pig serum and liver L-asparaginases. Purification of L-asparaginases by antibody precipitation.

Rabbits immunized with partially purified guinea pig serum and guinea pig liver L-asparaginases produced precipitating antibodies to the homologous enzymes. Antisera to guinea pig serum L-asparaginase precipitated the guinea pig liver and agouti serum L-asparaginases but did not crossreact with Escherichia coli B L-asparaginase. Conversely, antisera to guinea pig liver L-asparaginase precipitat...

Estimation for the Type-II Extreme Value Distribution Based on Progressive Type-II Censoring

In this paper, we discuss the statistical inference on the unknown parameters and reliability function of type-II extreme value (EVII) distribution when the observed data are progressively type-II censored. By applying EM algorithm, we obtain maximum likelihood estimates (MLEs). We also suggest approximate maximum likelihood estimators (AMLEs), which have explicit expressions. We provide Bayes ...

Isoaspartyl dipeptidase activity of plant-type asparaginases.

Recombinant plant-type asparaginases from the cyanobacteria Synechocystis sp. PCC (Pasteur culture collection) 6803 and Anabaena sp. PCC 7120, from Escherichia coli and from the plant Arabidopsis thaliana were expressed in E. coli with either an N-terminal or a C-terminal His tag, and purified. Although each of the four enzymes is encoded by a single gene, their mature forms consist of two prot...
